Purpose
TLC(Thin Layer Chromatography) have been used to confirm the radiochemical yield after synthesizing [18F]FDG. But TLC scanner dose not let us know other information such as chemical impurity or quantity of compound which did not have a radioactive decay. Many scientists reported that they used HPLC system for Q.C of [18F]FDG(Ryuji Nakao, et al. Nuclear Medicine and Biology 32(2005) 907- 912). So, we tried to compare two methods and which one was better.
Materials and Methods
We used AR2000 TLC Scanner(BIOSCAN co., USA), 321 pump, UV/VIS-155 detector(GILSON co., FRANCE) and Gabi RI detector(RAYTEST co., GERMANY). [18F]FDG was produced using SNUH Cyclotron facilities (TR13, EBCO co., CANADA). We purchased analytical column (Inertsil NH2., GL science, JAPAN) and FDG standard (ABX co., GERMANY).
Conclusion
Using TLC scanner, we got the results 15 min later. The results showed us several radio- compound peaks such as free [18F], [18F]FDG, [18F]TAG, and partially hydrolyzed [18F]TAG. In case of HPLC, we got the results by 18 min. The results showed us not only above all radio peaks but also UV peaks such as FDG, ClDG and other unidentified compound. But there was a pre-preparation to execute a HPLC method.
